What is the Snare complex?

A cascade of a trio of the SNARE proteins: synaptobrevin/VAMP, SNAP 25 (Synaptosomal Associated Protein), and Syntaxin (STNX) forms a complex with the ATPase NSF called SNARE complex.

The SNARE complex is involved in the trafficking of the synaptic vesicle containing neurotransmitters like acetylcholine to the presynaptic membrane. Fusion and exocytosis of the neurotransmitters occur, transmitting the signal from neuron to muscle for contraction.

How does BoNT-L inactivate the SNARE complex?

BoNT-L-peptide binds to SNAP-25 from the SNARE complex, preventing the fusion of the acetylcholine-filled vesicle with the plasmatic membrane. In its turn, it prevents the release of acetylcholine into the synaptic cleft between the nerve and muscle.
