Nonapeptide-1 is a synthetic biomimetic peptide that acts as an antagonist of α-MSH (melanin-stimulating hormone). It is found by screening methods using different parts of the α-MSH sequence and testing for their ability to block specific MC1-R receptors on melanocytes.

The most potent peptide was the Nonapeptide-1, which repeats a small part of the sequence (H-Met-Pro-D-Phe-Arg-D-Trp-Phe-Lys-Pro-Val-NH2) of α-MSH and occupies an MC1-R receptor, preventing its activation by the hormone and lessening melanin production by 33%. It prevents the activation of tyrosinase, a crucial enzyme involved in melanin synthesis.

Thanks to its antagonizing action, Nonapeptide-1 is an excellent skin-lightening agent without cytotoxicity or interference with melanocyte functions. It does not totally inhibit melanogenesis, which opposes the hyper-production of melanin pigment.
