Hexapeptide-42 is a synthetic peptide with sequence Ile–Gln–Ala–Cys–Arg–Gly–NH2 that activates the Caspase-14, a proteolytic enzyme expressed only in the epidermis and responsible for Filaggrin metabolism. Filaggrin is a protein that works as a source of NMFs (natural moisturizing factors), as its metabolites (amino acids) capture water molecules, providing a remarkable internal hydration effect in the epidermis. More importantly, Caspase-14 is involved in Profilaggrin (a precursor that contains 10-12 Filaggrin units) enzymatic conversion into Filaggrin during terminal differentiation of keratinocytes.
