Hexapeptide-42 is a synthetic peptide with sequence Ile–Gln–Ala–Cys–Arg–Gly–NH2 that activates the Caspase-14, a proteolytic enzyme expressed only in the epidermis and responsible for Filaggrin metabolism. In addition, it prevents UVB-induced DNA plerosis damage and stimulates repair mechanisms and cornification, preserving DNA integrity and improving skin barrier function.

Filaggrin is a protein that works as a source of NMFs (natural moisturizing factors), as its metabolites (amino acids) capture water molecules, providing a remarkable internal hydration effect in the epidermis. More importantly, Caspase-14 is involved in Profilaggrin (a precursor that contains 10-12 Filaggrin units) enzymatic conversion into Filaggrin during terminal differentiation of keratinocytes.

In short, Hexapeptide-42 has a perfect biomimetic target, Caspase-14, that improves skin hydration, DNA protection against UVB, and barrier function. Incorporated in skin care applications, Hexapeptide-42 acts as a multifunctional anti-aging agent fighting against photoaging, dehydration, and skin thinning.
